Expression of ¥â - glucuronidase Gene in Brassica napus Using Agrobacterium tumefaciens - mediated Transformation



Abstract
¥â-Glucuronidase (GUS) gene was introduced into Brassica napus cv. Velox by Agrobacterium tumefaciens-mediated transfornnation method, and the transformants were regenerated to whole plants. After 2 weeks of inoculation of the whole cotyledons at the cut petiole with Agrobacterium containing the pBI121 plant expression vector, the shoots developed directly from the cut edge of explants in contact with selection media which was supplemented with 15 §·/§¤ kanamycin. Transformation of the regenerated rapeseeds was confirmed by Southern blot analysis and expression analysis of GUS-gene. Each transformant contained one or two copies of GUS gene. Histochemical analysis was used to demonstrate the localization of GUS activity in tissues of transformed plants. Leaf and stem cross-sections showed the highest levels of GUS activity along inside and outside of the vascular system by showing strong blue staining. Transformation of direct shoot induction from cotyledon has been shown to be a simple and effective method for rapeseed.
